چکیده

The in uitm biosynthesis of acetylated human fetal hemoglobin (Hb FI) was investigated in umbilical cord blood from two normal newborns and in peripheral blood from five adults with different hematologic disorders accompanied by elevated levels of fetal hemoglobin. Hemoglobin biosynthesis was measured by [8HJleucine incorporation into hemoglobin components which were separated by chromatography on Bio Rex 70 cation-exchange resin. Following incubation of the samples for times ranging from 2% to 120 min, the specific radioactivities of newly synthesized Hb FI ranged from 2.1 to 3.9 times those corresponding values for the major fetal hemoglobin component (Hb F,) which is not acetylated. For both umbilical cord blood and the 8/3//3’ thalassemia blood, the ratio of specific radioactivities (Hb F1/Hb F,) decreased significantly during the initial 20 min of incubation. Pulse-chase experiments revealed that the newly synthesized Hb FI was stable. These results indicate that in the circulating reticulocytes of the newborn and in those of adults with elevated fetal hemoglobin, the rate of biosynthesis of acetylated fetal hemoglobin is higher than that expected from the amounts of this hemoglobin existing in the blood. Radiolabeled blood cells from an adult patient with 8/3//3’ thalassemia were separated into five zones by centrifugation in a density gradient. Although each of the five zones contained similar amounts of cellular hemoglobin, about 84% of the protein ‘H-radioactivity was present in the lightest (top) zone with the remainder distributed almost equally among the other zones. The Hb FI/Hb F, specific radioactivity ratio was about 1.5 in the lightest gradient zone and ranged from 11.1 to 18.4 in the four heaviest zones. These data suggest that much of the Hb F1 synthesis occurred in a separate population of erythroid cells heavier than, and probably more mature than, the cells responsible for most of the hemoglobin synthesis. This result may occur because of increased activity of an acetyltransferase enzyme which could serve as a marker for erythroid cell maturation.

Acetylated hemoglobins in feline blood.

The /3 chain amino terminus of one or more hemoglobins in the blood of members of the Felidae family is substituted with an acetyl group. Such acetylated hemoglobins occur together with nonacetylated components in variable proportions in blood of d8erent members of the family. In the lion, tiger, and snow leopard, the acetylated component comprises about 90% of the total hemoglobin; in the serv...

The LSD1 inhibitor RN-1 recapitulates the fetal pattern of hemoglobin synthesis in baboons (P. anubis).

Increased fetal hemoglobin levels lessen the severity of symptoms and increase the lifespan of patients with sickle cell disease. Hydroxyurea, the only drug currently approved for the treatment of sickle cell disease, is not effective in a large proportion of patients and therefore new pharmacological agents that increase fetal hemoglobin levels have long been sought. Recent studies identifying...

Change in Oxygen Absorption of Human Adult and Fetal Hemoglobin Due to 940 MHz Electromagnetic Field Radiation Exposure

The effects of electromagnetic fields (EMFs) radiation at the frequency of 940 MHz on the structure and function of human adult and fetal hemoglobin (HbA and HbF) were studied. After extraction and purification of HbA and HbF, the oxygen absorption values for exposed and unexposed HbA and HbF to EMF were compared. The slope of oxygen absorption curve for exposed HbA was increased while that for...

Hemoglobin synthesis in human BFU-E and CFU-E-derived erythroid colonies.

We have analyzed the synthesis of fetal hemoglobin (HbF) in the differentiated erythroid colonies produced by erythropoietin-responsive committed erythroid progenitors in plasma cultures of human marrow and peripheral blood. Two independent biochemical techniques. carboxymethyl-cellulose column chromatography of 3H-leucine-Iabeled globin. and polyacrylamide gel electrophoresis of 59Fe-labeled h...

Prenatal diagnosis of hemoglobinopathies: comparison of the results obtained by isoelectric focusing of hemoglobins and by chromatography of radioactive globin chains.

Isoelectric focusing (IEF) of hemoglobin was compared to the classical chromatography of labeled globin chains for 22 antenatal diagnoses of hemoglobinopathies: 11 for beta thalassemia, and 11 for sickle cell disease. In all cases, the two methods gave identical results. The diagnosis was confirmed after birth or abortion. Three fetuses homozygous for beta thalassemia and one homozygous for sic...
